While there is never any indication that they are something other than close friends in the books, in Harry Potter and the Deathly Hallows: Part 2, Neville, running on adrenaline, confesses that he has feelings for Luna and that he wants to tell her before the Battle of Hogwarts to Harry (Daniel Radcliffe) and Ginny. Luna is then seen sitting down next to Neville after the battle, teasing that they might get together later. However, Harry Potter's canon was extended via pottermore.com after the book series' completion, confirming that Neville and Luna both married other people in the book universe.

Luna is in her sixth year in the last Harry Potter book, with one more year to go. After Hogwarts, she became a Magizoologist, studying and caring for magical creatures. She also taught at Hogwarts for some time, one of the first professors of the Field Studies class, which allows students to travel from the school to observe and interact with magical creatures in nature.

In her personal life, Luna married another Magizoologist, Rolf Scamander, the grandson of Newt Scamander (played by Eddie Redmayne on screen), the protagonist of the Fantastic Beasts and Where to Find Them movies. Newt shared Rolf and Luna's profession and wrote the textbook that they both likely used while at Hogwarts; Newt's wife and Rolf's grandmother, Porpentina "Tina" Goldstein (Katherine Waterson), was an American auror.

Luna reportedly got married wearing rainbows and spangles, and a unicorn horn tiara, which Rita Skeeter's readers voted "The Most Hideous Outfit of the Year." Luna and Rolf had twin sons named Lorcan and Lysander. Harry and Ginny also named their daughter and youngest child Lily Luna Potter after Luna.

Neville, meanwhile, replaced Professor Sprout as the Herbology professor at Hogwarts sometime after graduation. He also became the Head of Gryffindor House, as the former head, Professor McGonagall, became the heistress of the school. Neville is also the godfather of Harry and Ginny's second son, Albus. Neville himself did not have any children but married Hannah Abbott, a side character in the Harry Potter series who is in the same year at Hogwarts as the main trio and Neville.

Hannah is in Hufflepuff while she is at Hogwarts, a member of Dumbledore's Army, and fights in the Battle of Hogwarts. It is briefly mentioned in Harry Potter and the Half-Blood Prince that she is pulled out of class because her mother had been found dead, murdered by Death Eaters. After school, she and Neville lived at the Leaky Cauldron, where Hannah was the landlady, but she later trained as a healer with an interest in becoming a nurse at Hogwarts.

Neville and Luna's later lives could technically be the same in the movie universe as in the books, as their being interested in each other when they are 17 doesn't mean they are going to get married. Harry Potter goes down this route with a lot of characters, which can be explained by many of them feeling like they will never have the same relationship with someone else after all they have been through together. Regardless, Neville and Luna led rich, happy lives after the series, and ended up married to interesting ing characters who were sadly never explored much.
