At the end of Marvel's Spider-Man 2, Peter Parker seemingly es the mantle to Miles, letting him take over the role of the primary Spider-Man in New York. However, that doesn't mean that he is the only Spider-Man in the city, and there is evidence that he will not be the only one in the next game. In fact, I believe that that will be far from the truth, and there will be more than just 2 Spider-heroes roaming New York in the sequel.
While Miles and Peter have starred in the past Spider-Man games, the roster of Spider-heroes is long and wide. Spider-Man's rogues gallery is just as deep, so sometimes it takes more than one hero to win the day. This can be seen in each of the past games. Even in the first game, Peter needed Miles and MJ's help to win, Miles got help from a number of people in his solo game, and both of them were needed in the last game. It's thinkable, then, that Miles will likely have a team-up in the next game.
Peter Parker Isn’t Gone Just Yet
Actor Confirms He Isn't Taking A Back Seat
In a recent interview for The Direct, actor Yuri Lowenthal confirmed that he is returning as Peter Parker and will not be sitting on the sidelines the whole time. While he might be focusing on the concerns for the Foundation, he hasn't lost his super-human abilities or his drive to help people as best he can. And, with Harry's condition growing worse, it's unlikely that he wouldn't step in as Spider-Man if it came down to it, particularly if the scene with Norman Osborne has some payoff.

While we haven't seen Green Goblin just yet, Norman referenced the G-serum, and Harry is left in a precarious position at the end of the game. It's likely that we see the emergence of the Green Goblin from one of them before the end of the game, sooner rather than later, and I would be shocked if this doesn't pull Peter back into Spider-Man duty. It's his best friend, and actions that have him or his father running around as a terrifying green monster will make him feel responsible for his friend's actions.
Two More Spider-Heroes Were Teased In Marvel’s Spider-Man 2
Multiple Cameos Suggest More Spider-Heroes Are Coming
Both Cindy Moon and Miguel O'Hara were teased in the game. Cindy Moon becomes Silk in the comics, and Miguel is Spider-Man 2099 and recently made a popular appearance in Across the Spider-Verse. Of the two, Cindy received the more direct reference, with Rio Morales having a direct connection to her at the end of the Marvel's Spider-Man 2. It would make sense if she came into the scene first, as Miles could potentially begin to mentor her in the same way Peter did for him.
Miguel was teased by a bartender named Delilah, who took some Spider-Bots and says, "If Miguel comes looking for these..." implying a connection to the futuristic Spider-Man. I don't think they would introduce time travel this early into the series, but it's entirely possible, particularly with the popularity of the Spider-Verse movies. They also haven't introduced any form of Ghost-Spider, but with Miles taking the lead position, this could change and, if dimension-hopping is possible, Miguel time-traveling wouldn't be out of the question either.
It's also worth pointing out that the games do not follow a specific timeline, as Peter and Miles were not originally in the same universe either, and the games have mixed various materials to create the world that which these two characters exist in. That being said, Gwen has not been teased and Miguel has, so of the two I would think Miguel might show up first, although not necessarily in Marvel's Spider-Man 3.
Miles Might Need Some Help With The Villains We Know About
There Are Lots Of Powerful Villains Teased To Come Soon
Miles is an extremely capable Spider-Man, as we have seen throughout his tenure in the past three games. However, there have been a lot of big villains teased, and he might end up needing a little help. Either Norman or Harry Osborne, or both, might be turning green soon, but we also saw Doctor Octavius coming up with his own plan at the end. There was also a Beetle DLC that was teased but eventually cut, which could make its way into the next game instead.

This Marvel's Spider-Man 2 Character Would've Been Perfect For DLC, So Why Isn't It Happening?
Carnage was a huge missed opportunity for Marvel's Spider-Man 2, and it's a big loss for players who were looking forward to playing the DLC.
But, perhaps, most dangerous of all, Carnage has been heavily teased in Marvel's Spider-Man 2. Carnage is a threat, and Miles might find himself needing some help to take Carnage down. Notably, Cindy has been a journalist in past iterations, which could get her invested in Carnage's story and a possible partnership with MJ. The Chameleon has also made an appearance in the previous game, which could cause a lot of confusion with his shape-shifting abilities, which would require strong teamwork from the Spider-heroes to defeat.
I'm convinced there will be a third addition to the team, and it makes sense that there would be 3 in the third game. I would bet on Cindy before anyone else, but that may not be true. Additionally, depending on how much Insomniac wants to lean into the Spider-Verse, there could be more than 3 with the reference to Miguel and Miles' connection to Ghost-Spider, although they might not all be playable. In my opinion, the more Spider-heroes the better in the next Marvel's Spider-Man game.
