There is no way Palpatine would have let an entire mint dissappear. Not only that, but there is strong evidence this mint has been missing since BEFORE the "Old Republic" with the references to "The Great Works". Unless they are going to use stasis shenanigans or introduce a new species with the longevity of Yoda, there is no way the Supervisor is human. It's probably just a Droid. They have made alot of effort to not show the supervisor, being revealed as a Droid would betray the people who would rebel against the lie they have been living under.
